Chemical Modification of Chitosan with 1-Aziridineethanol for the Increase of Amine Values

Abstract
To imporve transfection efficiency, we prepared 1-aziridineethanol-low molecular weight water-soluble chitosan(LMWSC) having the potential to form complex with DNA as a cationic polymer. 1-aziridineethanol-LMWSC(LMWSC-EA) was synthesized by ring opening reaction of 1-aziridineethanol with chitosan. The LMWSC-EA was characterized using H-NMR¡¿? 13 C-NMR. LMWSC-EA was complexed with plasmid DNA(pDNA) in various polymer/DNA(N/P) weight ratios, and the complex was identified using gel retardation assay. Therefore, LMWSC-EA will be useful in the development of gene carriers.
